This volume looks at the newest methods, materials, equipment, and technologies developed to study the cell biology of the pneumococcus. The chapters, which cover a wide range of topics, are organized into six parts: Part one discusses the cultivation of Streptococcus pneumoniae in vitro; Part Two talks about the microscopy techniques used to study the biology of Streptococcus pneumoniae and pneumococcal interactions with the host; Parts Three and Four describe the genetics, proteome, and proteomics of Streptococcus pneumoniae; Part Five explains in vitro and in vivo models of Streptococcus pneumoniae-host interactions; and Part Six looks at the factors of public health, epidemiology, and biostatistics. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Cutting-edge and authoritative, Streptococcus pneumoniae: Methods and Protocols is a valuable resource for both novice and expert researchers who want to expand their studies into new areas and new systems in this continuously evolving field.

Based on a workshop titled "Streptococcus pneumoniae: Molecular Biology and Mechanisms of Disease -- Update for the 1990s" held in September 1996 in Oeiras, Portugal, this volume contains some 40 contributions written by some 50 biochemists, molecular cmicrobiologists, geneticists, zoologists, pharmacologists, and pediatricians from 12 countries. Contributions are divided into six sections: an introductory segment addressing work to be done in the field and the disease's functional anatomy; the disease's chromosome structure, recombination, and cloning; capsule, cell wall, and virulence factors; Pneumococcal disease and

Streptococcus pneumoniae causes life-threatening invasive diseases in children and older adults. Although effective at reducing the incidence of disease, the two currently available vaccines against S. pneumoniae have significant limitations that a vaccine based on protein antigens may overcome. For this thesis I have investigated the role of S. pneumoniae ABC transporters during infection, and have assessed as potential vaccine candidates the lipoprotein components of two ABC transporters. Eleven ABC transporters were chosen for investigation of their role during infection,

Little is known regarding the role of Toll-like receptors (TLRs) in regulating protein- and polysaccharide-specific immunoglobulin (Ig) isotype production in response to an in vivo challenge with an extracellular bacterium. In this report we demonstrate that MyD88-/-, but not TLR2-/-, mice are markedly defective in their induction of multiple splenic proinflammatory cytokine- and chemokine-specific mRNAs after intraperitoneal (i.p.) challenge with heat-killed Streptococcus pneumoniae capsular type 14 (S. pneumoniae type 14). This is correlated with analogous responses in

Colonization of the pharynx by Streptococcus pneumoniae was studied in 185 in-hospital geriatric patients (median age 81 years) from 29 March 2011 to 22 June 2011. Swabs were plated on blood agar plates. Colonies with a morphology suggesting S. pneumoniae were further analyzed. Surprisingly, pneumococci were not found in any of the samples. Pneumococci chronically colonizing the pharynx of elderly people may be much rarer than previously thought and probably are not the source of pneumococcal pneumonia in old age....

Streptococcus pneumoniae is a major pathogen that causes Otitis Media infections and bacterial meningitis in children as well as community acquired pneumonia in adults. Clinical isolates of S. pneumoniae exhibiting resistance to Beta-lactam antibiotics are being isolated with increased frequency in many countries. Streptococcus pneumoniae strains resistant to Beta-lactam drugs have modified forms of penicillin-binding proteins that exhibit reduced affinity for binding to chemotherapeutic Beta-lactams. Penicillin binding proteins are membrane-bound enzymes that catalyze the terminal step in cell wall synthesis,
