✏Proteomics Biomedical and Pharmaceutical Applications Book Summary : Human biology has now entered into a phase of post-genomics and it might not be an exaggeration to say that the major outcome of the human genome sequencing has finally been to open the way to the exploration of the proteome-proteomics. Proteins are the functional output of genes and there are two main expected outcomes from human proteomics. The first is to discover new molecular markers for early diagnosis and profiling of pathologies. The second is to decipher the intracellular signaling pathways leading to the initiation and progression of pathologies, for the identification of new targets and the development of innovative therapeutic strategies. This is clearly a promising challenge that this book explores through a series of ongoing experiences and projects representative of the new era in which biology and medicine have now entered.

✏Low Abundance Proteome Discovery Book Summary : Low-Abundance Proteome Discovery addresses the most critical challenge in biomarker discovery and progress: the identification of low-abundance proteins. The book describes an original strategy developed by the authors that permits the detection of protein species typically found in very low abundance and that may yield valuable clues to future discoveries. Known as combinatorial peptide ligand libraries, these new methodologies are one of the hottest topics related to the study of proteomics and have applications in medical diagnostics, food quality, and plant analysis. The book is written for university and industry scientists starting proteomic studies of complex matrices (e.g., biological fluids, biopsies, recalcitrant plant tissues, foodstuff, and beverage analysis), researchers doing wet chemistry, and graduate-level students in the areas of analytical and biochemistry, biology, and genetics. Covers methodologies for enhancing the visibility of low-abundance proteins which, until now, has been the biggest challenge in biomarker progress Includes detailed protocols that address real-life needs in laboratory practice Addresses all applications, including human disease, food and beverage safety, and the discovery of new proteins/peptides of importance in nutraceutics Compiles the research and analytic protocols of the two scientists who are credited with the discovery of these landmark methodologies, also known as combinatorial peptide ligand libraries, for the identification of low-abundance proteins

✏Proteomics in Functional Genomics Book Summary : A wealth of information has accumulated over the last few years on the human genome. The new insights have completely changed the focus of protein analysis. It is no longer time-consuming analysis of unknown products, but rather selective identifications of individual forms, modifications and processings, and overall analysis of global protein outputs from cells and tissues in health and disease. This book gears to the rising need of sensitive, accurate, and fast separation and identification techniques in proteomics. It discusses current methodologies of modern protein analysis, from isolation and sample preparation, over analysis and identification, to final characterization. Several evaluations concentrate on the now productive approaches of two-dimensional gel electrophoresis and mass spectrometry, but alternative methods and further perspectives are also outlined. The book includes an overlook over current databases to connect protein analysis data with all available information,...

✏Separation Methods In Proteomics Book Summary : Driven by the widespread growth of proteomic practices, protein separation techniques have been refined to minimize variability, optimize particular applications, and adapt to user preferences in the analysis of proteins. Separation Methods in Proteomics provides a comprehensive examination of all major separation techniques for proteomics research. Written as a compilation of hands-on methods exemplified by the work of several recognized leaders in the field, this book may serve as a guide for selection of the optimal separation strategies to solve particular biological problems. Recent progress in the development of robust analytical techniques and instrumentation has created the need for good quality biological samples that are subject to analysis. Emphasizing the importance of sample preparation, the book explains how proteomes can be divided into smaller, less complicated “subproteomes" for individual analysis. It also highlights several hybrid approaches that take into account protein interactions. Including applications of the separation methods currently employed in proteomic analyses for both clinical and basic research, Separation Methods in Proteomics contains practical information that can enhance the current and future endeavors of scientists in proteomics, genomics, transcriptomics, biomarker discovery, and drug discovery.

✏Applications of Monolithic Column and Isotope Dimethylation Labeling in Shotgun Proteome Analysis Book Summary : In this thesis, the author outlines the development of new monolithic columns and isotope dimethyl labeling strategies and their applications in high-performance proteome analyses. Though different types of monolithic columns have been widely developed for chromatography and electrophoresis separation, their application in proteomics for complex peptide mixtures separation is still a challenge. The author discusses the preparation of new monolithic columns and optimization of chromatography separation capability to improve coverage and accuracy of proteome analysis. Further, the author describes a novel online multidimensional chromatography system combined with automated online isotope labeling, which significantly improves the throughput, sensitivity and accuracy of quantitative proteomics. In addition to the development of new technologies, the author investigates the proteome and phosphoproteome expression changes of clinical hepatocellular carcinoma tissues and the hippocampi of mice with Alzheimer’s disease. The work in this thesis has led to several publications in high-profile journals in the fields of analytical chemistry and proteome research.
