The ocean biomass, covering two-third of the earth, with huge unexplored natural product offers enormous scope and presents an effective alternative in natural product drug discovery. The uniqueness in oceanic mega-diversity is due to spatial as well as temporal competition along with unique habitat with extreme pressure, temperature, and saline conditions. As a result of this, marine organisms have adapted and evolved themselves successfully since centuries in these conditions by producing molecules which are unique in structures, biosynthesis, and function. This “chemical adaptations” is an excellent source of novel chemical entities which is absent in land-based organisms. The past decade has seen more than 10,000 compounds isolated from marine sources which have dramatically increased the number of preclinical anticancers drug under evaluation, and over 300 patents on bioactive natural products from marine sources were granted during this tenure. Efforts, in this direction, became more serious and focused with National Cancer Institute, USA taking a lead role. By collaborative interactions between pharmaceutical companies and research organization, numerous drug-like molecules with several of them having clinical and preclinical potential were discovered. Ecteinascidin-743/ET-743 from Caribbean tunicate and Didemnin and Aplidine from Aplidium albicans are some of the successful examples. Sterols and dietary fibers from seaweeds also hold immense potential. However, investigation of the marine floras chemical entities as drug-like molecule is still in its embryonic stage. (Family-Anacardiaceae). In phytochemical screenings with the crude extractives demonstrated the presence of alkaloids, steroids, tannins, saponins and gum. The antibacterial and antifungal activities of the crude extracts were evaluated by the disc diffusion method against 4 Gram positive and 7 Gram negative pathogenic bacteria and 7 fungi using Ciprofloxacin and Fluconazole as standards, respectively. In the screening, ethanol extract of L. grandis showed varying degrees of antibacterial activities than other two fractions with % zone of inhibition ranging from 46.09-48.42%, with strong activity against B. subtilis and S. aureus. On the other hand, the ethanol extract of plant also revealed mild to moderate antifungal activity with highest zone of inhibition of 20.5mm against B. dermatitidis. The MICs of the extracts were found to be 15.625-250μg/ml for both bacteria and fungal species used in the screening. ✏Phytochemical Investigation of the Medicinal Plant Taxodium Distichum and Library Screening of Thalictrum Alkaloids for New Antileishmanial Drug Leads Book Summary : Leishmaniasis is an infectious disease caused by blood-borne parasites from the genus Leishmania. The therapeutic options currently available for this potentially deadly and disfiguring disease are limited in their number and availability to at-risk populations, and also cause severe toxicity and side effects. An increasing incidence of drug resistance has also been developing among clinical strains of Leishmania parasites. There is thus a great need for the discovery and development of new antileishmanial agents. Natural products research has historically been a successful avenue for the discovery of new drugs and lead molecules for medicinal chemistry optimization efforts. This may be because natural products tend to be structurally complex and suited for interaction with various drug targets in biological systems. Through collaborative studies on the in vitro antileishmanial activities of plants growing in Ohio, the extract produced from bald cypress (Taxodium distichum) cones was found to be active and was selected for phytochemical investigation. Additionally, a chemically diverse set of 234 molecules in a library of previously isolated plant natural products was tested against L. donovani, which is responsible for the fatal (visceral) manifestation of leishmaniasis.

✏Phytochemical and Biochemometric Evaluation of the Alaskan Anti tb Ethnobotanical Book Summary : "The present study demonstrates new bio-analytical approaches for the evaluation of ethnobotanicals with a focus on the chemical and anti-TB biological analysis of Oplopanax horridus Michix (Devil's Club). Anti-TB activity analysis of crude extract and fractions of O. horridus resulted in the confirmation of a synergistic interaction among active anti-TB fractions with maximum enhancement of two fold for the recombined fractions. The combination of CCC, MABA, and the isobole method was proven to be effective for the evaluation of synergy in a complex mixture of chemicals. In vivo experiments of concentrated active fraction did not show toxicity or significant activity at 1,000 mg/kg dose level. Chapter 4 details with the bioassay-guided fractionation of O. horridus, where the isolation of five compounds including two new sesquiterpenes: 3,10-epoxy-11-hydroxynerolid-6-ene (1) and 3S,6R,7S,10R-cis-6,11-dihydroxy-7,10-epoxynerolidol (2), and two known active compounds: oplopandiol (3) and falcarindiol (4) is demonstrated. A newly developed Gradient-array CCC method was proven to be an effective and rapid separation method for the crude extract with little chemical overlap. Iteration of 1H NMR spectra revealed the full spin system analysis of compounds 2 and 5. It was demonstrated for compound 5 that it is feasible to dereplicate an isolated compound at the sub-mg level, including its stereochemistry, with only with MS and 1H NMR spectra with an aid of iteration software. A new model to evaluate medicinal plant preparations in a quantitative manner without isolating active constituents was also established. Named biochemometrics, the model allowed successful identification of the four known active compounds, while excluding the inactive compounds from the positive matrix. Also, it identified 24 additional compounds in the active fractions by mathematically correlating the biological and chemical information with an aid of MS based dereplication. Consistencies in skeletal structures were observed among compounds identified by MS fragmentation patterns. Hence, biochemometrics was found to be effective tool to identify classes of compounds that exhibit high potential for anti-TB drug leads by consistently detecting compounds with the same skeletons at various KD and retention times. The parameters involved in biochemometrics analysis were evaluated to improve the accuracy of the analysis. Extract preparation of Oplopanax horridus constitutes an example of an ethnobotanical in which active constituents concertedly work together to exhibit anti-TB activity. These findings lend support to the recent trend to recognize the fact that the biological effects of herbals are often greater at the stage of crude extract preparation when compared to the pure (active) compound level. The present study confirms this by proving both active principles and synergy, and by developing biochemometrics as a new tool for their integrated phytochemical analysis.
