✏Novel Roles of RelB in Neuroinflammation Book Summary : Neuroinflammation is hallmark of almost all central nervous system diseases. Along with infiltrating immune cells, resident glial cells of the central nervous system play a critical role in modulating inflammation and thus regulating disease severity. The NF-kB family of transcription factors are major transcriptional regulators of inflammation. RelA (p65) is the most well studied NF-kB family member; it has been well established that p65 functions to increase cytokine transcription in a variety of disease models. However, other NF-kB family members, namely RelB, have not been studied extensively as regulators of neuroinflammation. This report describes the both the function of RelB in glial cells during sterile neuroinflammation and during auto-immune neurological diseases such as multiple sclerosis. In the first part of this project we focused on studying astrocytes, as these glial cells are key regulators of homeostasis in the brain. We have established that primary human astrocytes exposed to pro-inflammatory cytokines such as IL-1 develop days-long "cytokine-tolerance " to subsequent cytokine activation. This adaptive component provides "memory " or "imprinting " of astrocytes, which resembles endotoxin-induced tolerance observed in macrophages. Mechanistically, we found that "cytokine-tolerance " in astrocytes depends on RelB. Although IL-1 initially activates the classical NF-kB protein p65, it subsequently induces RelB expression. Upon re-exposure to IL-1, the excess of RelB containing complexes occupy NF-kB binding sites on DNA diminish further expression of cytokines, and therefore limit inflammatory responses. We further confirmed our findings in an LPS-induced model of neuroinflammation, demonstrating that astrocytic RelB knockout results in increased cytokine expression in the brain. In the second part of this project, we investigated the role of glial cell derived RelB in experimental autoimmune encephalomyelitis (EAE), a mouse model of multiple sclerosis. In contrast to the LPS- induced model of neuroinflammation, astrocytic RelB had no effect on disease severity. However, we discovered oligodendrocyte-derived RelB functions to increase disease severity possibly by increasing cell death. These two projects demonstrate the crucial functions of glial cell derived RelB in regulating neuroinflammatory diseases.

Recent evidence has linked MMPs to conditions like ischemia, multiple sclerosis, Alzheimer's disease and suggested that, together with their role in the degradation of extracellular macromolecules, MMPs could work as important signalling molecules from injured neurons to the microglia. Thus, MMP-3 has been shown to induce the release of pro-inflammatory cytokines from microglia via activation of ERK and NF-kB-dependent pathways. Increasing evidence highlights the importance of a balanced cross-talk between neurons and non-neuronal cells and indicates that the presence of reactive astrocytes, the activation of microglia and the release of inflammatory mediators may contribute to the progression of many central nervous system disorders, independently of the nature of the primary pathogenic event. However, many aspects still remain controversial and much more effort is needed to understand the role of neuroinflammatory mediators and processes in these conditions. ✏Role of the Neuroinflammation in the Degree of Spinal Cord Injury New Therapeutic Strategies Book Summary : A case of spinal cord injury (SCI) is defined as the occurrence of an acute traumatic lesion of neural elements in the spinal canal (spinal cord and cauda equina), resulting in temporary or permanent sensory and/or motor deficit. Most studies on traumatic SCI show a bimodal age distribution, with a first peak in young adulthood and a second peak in older adults. Spinal cord trauma activates a cascade of events that exacerbates the damage such as activation of inflammatory process that determinates cytokine and chemokine production and that generates reduction in functional recovery resulting in necrosis or apoptosis of neurons. However, the precise mechanism of SCI-induced inflammatory response remains not fully understood at present. Current strategy to treat damage to the spinal cord is limited, only the treatment with methylprednisolone (MP), if administered in excessive dose during the acute phase of the damage, could ameliorate patients with severe SCI. However, associated to the beneficial effects, there are growing evidence that high-dose of MP is correlated to increased risk of infections, pneumonia and gastrointestinal bleeding. Therefore, there is a necessity to develop new therapies to treat SCI; one of these is to selectively reduce inflammation that possess unique role in the processes of injury and recovery.
