BRPF2 and BRPF3 are paralogous to BRPF1, so these three human regulators form a subgroup within the bromodomain superfamily, composed of 39 other members. Recently we have reported that mouse Brpf1 plays a critical role in pre- and post-natal development and found that total Brpf1 knockout mice display embryonic lethality at E9.5. Other groups reported earlier that Brpf2 is required for erythropoiesis and its deficiency causes mouse embryonic lethality at E15.5, but the role that BRPF3 plays in vitro and in vivo remained elusive until two years ago when we reported its functions (as shown in Chapter II of this thesis). We took molecular and mouse genetic approaches to investigate the functions of BRPF3. The results showed that endogenous BRPF3 preferentially forms a tetrameric complex with HBO1 in mammalian cells and mouse Brpf3 disruption results in no obvious gross phenotypes. The findings indicate that BRPF3 is not required for mouse development and survival, thereby distinguishing it from its paralogs. Consistent with our previous studies that Brpf1 is critical for mouse cerebral development, we have recently reported heterozygous germline human BRPF1 mutations in over ten patients (as shown in Chapter III of this thesis). Patients with BRPF1 mutations are associated with a novel developmental disorder characteristic intellectual disability, ptosis and other symptoms due to defective histone H3 acetylation at lysine 23, an abundant but poorly characterized histone mark. This new study highlights the importance of the epigenetic regulator BRPF1 in human development. Moreover, acetylation-like acylations, such as propionylation and crotonylation, have recently been identified as novel histone modifications, but their links to human diseases remain to be established. Two important questions are whether histone acetyltransferase MOZ or MORF catalyzes these novel modifications and how BRPF1 may regulate these modifications. Related to these questions, we presented, in Chapter IV of this thesis, a complete but unpublished study that the MOZ-BRPF1 complex catalyzes propionylation of histone H3 at lysine 23 (H3K23) in vitro and in vivo. The modification is dramatically decreased in Brpf1 knockout mouse MEFs and tissues and in lymphoblastoid cell lines (LCLs) derived from human patients with the aforementioned developmental disorder. Interestingly, deacetylase inhibitors rescue the loss of H3K23 propionylation in the patients’ LCLs, suggesting the potential clinic application for treating patients with this and similar developmental disorders. Furthermore, we found that acetylation and propionylation of H3K23 increase significantly in ten cancer cell lines, including MCF7 (breast cancer), H1299 (lung cancer) and PC-3 (prostate cancer). The BRPF1-selective inhibitor PFI-4 and the MOZ-specific inhibitor WM-8014 effectively block these two acylations in the cancer cell lines, thereby suggesting potential clinical applications of these findings. Taken together, BRPF1-controlled histone H3 acylations at lysine 23 are required for normal development, but hyper-acylations might contribute to cancer initiation and progression. These findings thus uncover the important roles of BRPF1 in mouse development and human disease"--

Thus systematic integration of genetic, epigenetic and epidemiological approaches will contribute to improving our understanding of the molecular mechanisms underlying disease phenotypes.
