It is biocompatible, biodegradable, mucoadhesive, soluble in acidic-solutions, nontoxic and nonallergenic. The main drawback of chitosan in pharmaceutical procedures is its low solubility in physiological medium. Chitosan shows physicochemical characteristics that allow it to interact with a wide variety of molecules. This is of particular interest when increasing the solubility of poor water-soluble drugs. For this purpose, chitosan can be used in oral, nasal and ocular routes. In order to modulate drug release rate and achieve a proper drug delivery in physiological medium, some parameters can be modified when solid dispersions or nanoparticles (NPs) based on chitosan are being designed. In case of nanoparticles, chitosan can be used as the main component or as a modifying agent. In order to optimize drug loading and drug delivery, response surface methodology (RSM) is an interesting tool usually underestimated in the pharmaceutical field, which allows us to optimize the parameters involved in the process simultaneously and not by different steps, which usually lead to mistakes.

Rheology analysis shows that the gelation temperature of NSHBC hydrogels is 3.8 °C higher than that of hydroxybutyl chitosan (HBC) hydrogels. A model drug bovine serum albumin (BSA) is successfully loaded and released. NSHBC hydrogels show excellent pH sensitivity drug release behaviors. After incubation for 24 h, 93.7% of BSA is released from NSHBC hydrogels in phosphate buffer saline (PBS) (pH 7.4), which is significantly greater than that of 24.6% at pH 3.0. In contrast, the release rate of BSA from HBC is about 70.0% at pH 3.0 and 7.4. Thus, these novel hydrogels have the prominent merits of high adaptability to soluble drugs and pH sensitivity triggered release, indicating that NSHBC hydrogels have promising applications in oral drug delivery. Abstract : Thermo/pH‐sensitive N ‐succinyl hydroxybutyl chitosan (NSHBC) hydrogels are first prepared and investigated for drug delivery. ✏An Overview of Chitosan Xanthan Gum Matrices as Controlled Release Drug Carriers Book Summary : Naturally occurring polysaccharides and/or their chemically modified derivatives have been widely investigated in relation to their use as components of controlled release systems for drug delivery. The aforementioned is due, in part, to their distinct properties such as abundant availability and biocompatibility as well as environmental and economic advantages. Chitosan (CS) and xanthan gum (XG) based matrices have received growing scientific/pharmaceutical interest as oral controlled release drug carriers. Herein, recent advances spanning the last two decades in CS-XG based drug delivery systems are reviewed with the emphasis being on oral tablet formulations, due to their versatility as pharmaceutical dosage forms. The mechanism of interaction between CS and XG, by means of computational and experimental approaches, is scrutinized. Results obtained from the literature establish the possibility of fabricating a controlled release drug delivery system based on CS and XG matrices. This can be achieved by monitoring and manipulating the physiochemical properties of the two polymers as well as the experimental variables affecting their drug retardation efficiency, without the need to employ special equipment or sophisticated experimental techniques/methodologies.
